Geez, there I was trying to figure out if the one above was a video game but it was Bleach! ...I do not know Maniak's one either.
Guy gets cursed to find stones to help him remove it, but he seems to get dragged into helping a doctor cure a bunch of people, yet they both go on hunting with a lady and her cat, bodyguard, cleric...where'd all these people and unrelated tasks come from?! He just wants to remove his curse dang it!
(note: I know it isn't really a curse)

Outcast orphan somehow gets roped into a terrorist's plot to harvest all of mankind's energy for a bunch of moon people when he just wants peace and friendship.

I want to say yes to that, and now can't believe I unintentionally managed to get InuYasha out of that too, with that being Miroku.
Was actually meant to be Therion's side of Octopath with him meeting Alfyn then by chance H'annit with either a monster or Olberic, then Ophilia with "all these people" being the others plus Primrose and Ophilia's followers.

I woke up thinking about what I had put last night and hated it being so simple, so I came to change it to something better.
I believe yours is Mass Effect, but don't know which one, I assume the first.
Two brothers head on a journey to retrieve the literal voice of royalty with aid from a mortal enemy who becomes their mortal enemy again regardless, twice, but both times slightly different.
